abstract Described is a method for selecting cells with a zero level of fucose, applicable as host cells for the expression of recombinant proteins. This method involves: 1) introducing genetic mutations into a population of CHO cells by contacting these cells with methotrexate (MTX), 2) contacting said population of CHO cells containing mutant cells with a non-toxic fucose-binding substance for a time that makes it possible to carry out binding a fucose-binding agent to a fucose group on the cell membrane of a cell population where a period of time does not allow cell killing to take place; and 3) extracting from this population of cells containing mutant cells, a subpopulation of cells that bind to a fucose-binding agent, thus selecting cells useful as host cells for expression of recombinant proteins, the selected cells have zero fucose content. Cells and cell lines useful as host cells for the expression of recombinant proteins are also described.
