abstract The present invention relates to an amide and urea derivative of heteroaryl substituted diazatricycloalkanes of the formula (I), as defined in the description, to pharmaceutical compositions comprising the compounds, to methods for preparing the compounds and to methods of treatment using the compounds. More specifically, treatment methods include modulating the activity of the α7 nAChR subtype by administering one or more compounds to treat or prevent disorders mediated by the α7 nAChR subtype. Diazatricycloalkanes typically consist of 1-azabicyclooctane fused to a pyrrolidine ring. Substitutional heteroaryl groups are 5- or 6-membered ring heteroaromatic compounds, such as 3-pyridinyl and 5-pyrimidinyl fragments, which are attached directly to the diazatricycloalkane. The secondary nitrogen of the pyrrolidine moiety is a substituted arylcarbonyl (derivative of the amide type) or arylaminocarbonyl (N-arylcarbamoyl) (derivative of the urea type) group. The compounds are medicinal in therapeutic applications requiring selective interaction on certain nAChR subtypes. That is, the compounds modulate the activity of certain nAChR subtypes, especially the α7 nAChR subtype, and have no noticeable activity with respect to muscarinic receptors. Variants of compounds labeled with radioactive isotopes can be used in diagnostic methods.
