abstract | The invention relates to a new method for the biochemical synthesis of 1,4-butanediamine in a microorganism having an increased level of orinine decarboxylase activity (increased ODC activity) as compared to the native level of ornithine decarboxylase, where an increased activity of ODC is obtained by overexpression of a gene encoding ornithine decarboxylase with an increased transcriptative pattern. efficiency and where 1,4-butanediamine produced in the microorganism is secreted into the fermentation medium and recovered from the fermentation medium. In preferred embodiments, the enhanced enzyme activity is obtained by overexpression of either (i) the speA gene, encoding arginine decarboxylase, and the speB gene, encoding agmatinase; (ii) the speA gene, encoding arginine decarboxylase, and the aguA gene, encoding agmatinimine hydrolase, and the aguB gene, encoding N-carbamoylputresinamide hydrolase, and optionally the speB gene, encoding agmatinase. The invention also relates to vectors, plasmids and hosts, carrying an increased level of activity of one or more of the above mentioned enzymatic activities. |
