Compounds, methods of use and methods for producing complement factor D inhibitors are provided, including a compound of formula I or a pharmaceutically acceptable salt or composition thereof. The inhibitors described in the present invention have a directed effect on factor D and inhibit or regulate the cascade of complement reactions due to its early and important role in an alternative complement activation pathway and reduce the ability of factor D to modulate classical and lectin complement activation pathways. The factor D inhibitors described in the invention are able to reduce the excess activation of complement, which has been associated with certain autoimmune, inflammatory and neurodegenerative diseases, as well as ischemic reperfusion injury and cancer. |
