abstract The invention relates to a method for producing a protein scaffold that specifically binds to a target molecule. The method includes contacting a library of type III fibronectin domains (FN3) with a diversified alternative surface of C-CD-F-FG formed by beta-strand C, loop CD, beta-strand F and loop FG, with the target molecule and the selection of the protein frame , which specifically binds to a target molecule with a given affinity, where the library of FN3 domains with a diversified alternative surface of C-CD-F-FG is formed by introducing mutations in the reference polypeptide of the FN3 domain with the amino acid sequence of SEQ ID NO: 27, where the mutations include L32 residues , Q34 and Q36 of the beta strand C, provided that S30 is not mutated, residues T68, S70 and Y72 of the beta strand F, provided that the residue E66 is not mutated, residues S38, E39, K40 and V41 of the CD loop, provided that not residues G42 and E43 are mutated, and residues H78, R79 and N81 of the FG loop, provided that residues K75, G76, G77 and S80 are not mutated, where the residue numbering corresponds to SEQ ID NO: 27.
