abstract | The invention relates to the unexpected discovery that modification of the structure of a glycan on a human SAP polypeptide may increase the biological activity of this SAP polypeptide relative to a corresponding wild-type SAP sample isolated from human serum. This discovery provides glycosylated human SAP polypeptides containing an N-linked oligosaccharide chain in which all sialylated branches of the oligosaccharide chain end with an α2.3-linked portion of sialic acid, and the oligosaccharide chain is free of α2,6-linked parts of sialic acid, as well as having IC inhibition of monocyte differentiation into fibroblasts in vitro, which is less than the corresponding wild-type SAP sample isolated from human serum. The invention also relates to pharmaceutical preparations containing such SAP polypeptides, methods of producing such SAP polypeptides, CHO cells containing such polypeptides, and the use of such SAP polypeptides. |
