abstract |
In vitro isolation or assembly of the Streptococcus thermophilus CRISPR3 / Cas system Cas9-crRNA complex and using for cleavage of DNA carrying the nucleotide sequence complementary to the crRNA and adjacent to the protospacer motif. Methods for site-specific modification of a target DNA molecule in vitro or in vivo using an RNA-directed DNA endonuclease containing RNA sequences and at least one of the motives of the active RuvC site and the motives of the active HNH site; to convert a Cas9 polypeptide to nikkazu, splitting one strand of double-stranded DNA by inactivating one of the active sites (RuvC or HNH) in the polypeptide using at least one point mutation; to assemble an active polyribonucleotide polypeptide complex in vivo or in vitro and to reprogram the specificity of the Cas9-crRNA complex in vitro and using a cassette containing a single repeat-spacer-repeat unit. |
