abstract | The invention relates to a method for determining the sensitivity of microorganisms to chemical agents or antibiotics, namely, that microorganisms are cultivated in a liquid nutrient medium with a chemical agent or antibiotic, then the medium is treated in such a way that cell lysis and distribution of fragments of microorganisms by volume of the medium can occur, and then in the supernatant that does not contain microorganisms, measure the content of protein components and compare the values with the control or standard samples. The method involves adding a certain number of microbial cells to a liquid nutrient medium, to which a reinforcing agent specific for a particular group (genus or family) of microorganisms is added. The liquid nutrient medium containing microorganisms is preincubated, and then after adding a certain amount of antibiotic, it is again incubated. The cells are then removed from the incubation medium, obtaining the supernatant. The supernatant proteins are extracted by adding alcohol, followed by isolation of the proteins. Protein precipitate is used to obtain the mass spectrum used for comparison with standard mass spectra. |
