abstract |
The invention relates to a method for producing a slow-release estradiol composition comprising (a) preparing particles consisting of 17-β-estradiol and cholesterol in a 1: 1 ratio between 17-β-estradiol and cholesterol, where each of them is in amorphous or polymorphic form; (b) exposure of the atmosphere to the particles from step (a) at a relative humidity of 25% or less for 24 hours; (c) exposure of an atmosphere saturated with acetone and water to the particles from step (b) for 72 hours at 60 ° C; (d) heating the particles from step (c) at 45 ° C for 42 hours, while the dissolution rate of 17-β-estradiol (SRE) from the particles obtained in step (d) is less than 6% when measured in water a solution of 0.3% wt./about. polyoxyethylene sorbitan monooleate (Tween80®) and water purified according to USP for 24 hours at 37 ° C and standard pressure. The composition of slow release, obtained by the method according to the present invention, gives dual advantages to the composition with a low dose of estradiol with a low-frequency administration scheme. The composition may be administered parenterally once a month or at a lower frequency. |
