abstract The invention relates to a method for producing at least 1 × 10 viral particles (VP) / ml of E1-deficient serotype 26 adenovirus (rAd26), the ratio of the absolute number of virus particles to infectious particles VP / IU is less than 20: 1, and the method includes a) cultivating PER.C6 cells in suspension using a perfusion system; b) infection of these cells by rAd26 at a density of 10 × 10 to 16 × 10 viable cells / ml; c) further culturing infected cells using a perfusion system to propagate said rAd.26 until a concentration of at least 1 × 10 viral VP / ml particles is reached and d) collecting viral particles of said rAd26. The invention also relates to the use of a bioreactor in said method, where said bioreactor has a working volume of from 2 to 1000 liters and contains culture medium, PER.C6 cells and rAd26 virus particles at a concentration of at least 1 × 10VP / ml.
