titleOfInvention METHOD OF OBTAINING H-CSF HUMAN
abstract The invention relates to a method for producing granulocyte colony-stimulating factor (G-CSF) with a high volumetric yield (5.8-8.3 g / l) under non-cyclic cultivation with feeding of bacterial cells transformed with a plasmid with a human G-CSF gene, in media containing 5-10 g / l of thiamine, 60-180 mM of potassium ions, as well as 60-300 mM of sodium ions or 150-250 mM of magnesium ions. Growing and increasing the biomass of these cells is carried out until the optical density of the medium is approximately 50 units (at a wavelength of 600 nm). Next, isopropyl thiogalactoside IPTG is added to induce the expression and production of G-CSF. The method allows to obtain G-CSF with a high volumetric yield due to the maintenance of high stability of the plasmid using a simple and stable process.
