abstract | The invention relates to a method for producing a covalent polyethylene glycol conjugate with interferon, characterized in that a) a free hydroxyl group of monomethoxy polyethylene glycol is acylated with nitrobenzoic acid halide in a halogenated organic solvent in the presence of an organic base; b) the nitro group of the nitrobenzoic acid methoxy polyethylene glycol ester obtained is reduced with a salt of the d-element 4 of the period IV-VI group to a low degree in an aqueous or aqueous-organic medium with a pH from 1.0 to 6.5 or in a polar organic solvent; c) aminobenzoic acid methoxypolyethyleneglycol ether obtained is diazotized with alkali or alkaline earth metal nitrite in an aqueous or aqueous-organic medium or organic nitrite in a polar organic solvent medium, followed by removal of excess nitrite and obtaining an activated pegylating agent; d) the activated pegylating agent is introduced into the azo-coupling reaction with interferon in an aqueous or aqueous-organic medium with a pH from 7.0 to 10.0 and e) the mixture of pegylation products is separated by ion-exchange chromatography to produce mono-interglylated interferon. A method of preparing an activated PEG agent is also provided. EFFECT: expansion of the spectrum of interferons with an increased circulation time in the bloodstream. |
