abstract | The invention relates to a method for producing a covalent conjugate of polyethylene glycol with erythropoietin, characterized in that: a) the free hydroxyl group of monomethoxy polyethylene glycol is acylated with nitrobenzoic acid halide in anhydrous pyridine; b) the nitro group of the nitrobenzoic acid methoxy polyethylene glycol ester obtained is reduced with a chromium (II) salt; c) aminobenzoic acid methoxypolyethyleneglycol ether obtained is diazotized with alkali or alkaline earth metal nitrite in an aqueous or aqueous-organic medium or organic nitrite in a polar organic solvent medium, followed by removal of excess nitrite and obtaining an activated pegylating agent; d) the activated pegylating agent is introduced into the azo coupling reaction with erythropoietin in an aqueous or aqueous-organic medium with a pH from 7.0 to 9.5; e) a mixture of pegylation products is separated by ion exchange chromatography to obtain mono-pegylated erythropoietin. Technical result: expansion of the spectrum of erythropoietin with an increased circulation time in the bloodstream. |
