The invention relates to methods for increasing the secretion of a protein of interest (POI) from a eukaryotic cell, providing for the co-expression of POI and at least one protein that enhances the secretion of the protein, wherein said enhancing (enhancer) protein is selected from the group consisting of BMH2, BFR2, COG6, COY1, CUP5, IMH1, KIN2, SEC31, SSA4, SSE1, as well as in addition to the yeast promoter sequence, in particular the promoter sequence of the P. pastoris PET9 gene, which, under comparable conditions, has an increased promoter activity relative to the promoter sequence of the GAP protein. In addition, the invention relates to an expression vector containing such a promoter sequence and to the use of such an expression vector for expressing POI in a host cell, as well as novel promoter gene sequences from P. pastoris that are useful for expressing POI in yeast. |
