abstract | The invention relates to the field of clinical biochemistry, biotechnology and medicine. Describes how to detect permissible for human and animal doses of pesticides based on changes in the activity of pancreatic phospholipase A in the presence of pesticide (experience) and without it (control) in relation to phosphatidylcholine in the form of artificial lipid membranes (liposomes) by determining the content of hydrolysis products using thin-layer chromatography. For experimental and control samples on the content of phosphorus in phospholipids and their lys derivatives, determine the degree of hydrolysis of the substrate, which characterizes the change in enzyme activity under the influence of a pesticide. The dose of the pesticide is determined as safe, provided that the activity of the enzyme changes in its presence within the range of not more than 20% of the control. The claimed method on a small amount of available raw materials (10-100 μg of pesticide) without using laboratory animals allows the relative degree of hydrolysis of liposomes, as a model of the cell membrane, to measure phospholipase A activity compared with the control value as an indicator of cell wall destruction during digestion in the presence of the tested pesticide and without it. |
