abstract | The present invention provides methods for determining the amount of nucleic acid in a sample. The methods described utilize the ability to disrupt and redirect PCR and the ability to physically pair nucleic acid molecules in a sample that have a reference sequence with nucleic acid molecules in the sample that have a target sequence. PCR redirection makes it possible to use partial amplification as a preparatory step for other methods within the same tube. Pairing may result in the presence of an unpaired target sequence or an unpaired reference sequence indicating a difference in the amount of the target sequence compared to the amount of the reference sequence. The methods of the invention are widely applicable for determining differences in the amount of nucleic acids in diagnostics and research. |
