abstract SUBSTANCE: proposed is a method for in vitro transcription of mRNA and/or translation of polypeptides, including carrying out the synthesis of the mentioned mRNA and/or polypeptides in a cell-free reaction mixture containing an antifoam additive. A reaction mixture for cell-free synthesis of biological macromolecules containing an antifoaming agent can be used to carry out the reaction on a larger scale, for example, in volumes greater than at least 15 μl. The reactions can be carried out in a variety of reactors known in the industry, including stirred reactors, bubble column reactors, and the like.
