The invention relates to the discovery of new soluble in neutral conditions soluble hyaluronidase glycoproteins (sHASEGP), methods for their preparation and their use to facilitate the introduction of other molecules or to remove pathologies associated with glycosaminoglycans. Minimal active polypeptide domains of soluble, active under neutral conditions sHASEGP are described, including the sugar parts of the molecule associated with asparagine, which are necessary for the functioning of the hyaluronidase domain active under neutral conditions. Aminobenzene leader peptides that enhance sHASEGP secretion are included. The invention further introduces sialylated and PEGylated forms of recombinant sHASEGP in order to increase stability and improve serum pharmacokinetics compared to the natural enzymes produced in slaughterhouses. The following describes suitable kits containing essentially purified recombinant glycoprotein sHASEGP derived from eukaryotic cells that produce proper glycosylation, which is necessary for its optimal activity. |
