The application describes a new method of controlling sialic acid present in the oligosaccharide side chain of the glycoprotein produced by the culture of mammalian cells by changing the specific productivity of the specified culture, which is carried out by varying the cell culture medium. For carrying out the invention, during the transition phase, the osmolality of the cell culture, the concentration of the enhancer of transcription and the temperature of cultivation are established and maintained at the stage of glycoprotein production. In addition, the invention relates to the production of a protein consisting of a soluble form of human tumor necrosis type 1 and immunoglobulin G, with an increased or decreased amount of sialic acid in the oligosaccharide side chain, as well as preparations and therapeutic compositions containing the stated glycoproteins. |
