abstract |
The invention concerns a process for preparing viruses and viral vectors by the transfection of mammal cells with complexes of viral DNA and a polycation, which is optionally conjugated with a ligand for the target cell, in the presence of an endosomolytic agent. The process affords particular advantages for the production of recombinant retroviruses, adenoviruses and adeno-associated viruses in the cell, permitting high virus titres in packing cell lines and in primary cells. The area of application is above all gene therapy. |
