abstract | A method for direct qualitative and quantitative detection of mutations in biopolymers (e.g. NDA, RNA) is claimed comprising (i) prodn. of specifically lablelled prods. by an allele specific PCR (ii) sepg. (i) by solid phase techniques, (iii) incubating (ii) with enzymatically labelled antibodies (ABs), and (iv) detection of (iii) by chemiluminescence. Pref., the primer for the opposite strand is biotin-labelled and the mutant primer is labelled with fluorescein or vice versa. USE/ADVANTAGE - The method is more sensitive and cheaper than known methods (1 mutant can be detected in a pool of 1000 DNA probes). The PCR prods. from a reaction which is only specific for one allele are coupled to a solid phase and indirectly detected by the labelled primer. This is in contrast to prior art methods where PCR prods. are not detectable after the first PCR reaction. |
