abstract The prepn. of (3R,5S)-3,5,6-trihydroxy hexanoic acid isopropylidene ketal derivs. of formula (I) comprises (a) converting (4S,6S)-7-benzyloxy -4,6-dihydroxy hept-1-ene of formula (II) into a (4S,6S)-7-benzyloxy 4,6-dihydroxyhept-1-ene isopropylidene ketal of formula (III); (b) oxidatively removing the double bond from (IV) with a NaIO4/KMnO4 system to give a (3R,5S)-6-benzyloxy-3,5-dihydroxyhexanoic acid isopropylidene ketal of formula (V); (c) converting (V) into a (3R,5S)-6-benzyloxy 3,5-dihydroxyhexanoic acid isopropylidene ketal ester of formula (VI); and (d) removing the benzyl gp. to give the alcohol (I). R2 and R3= 1-6C alkyl, or together form (CH2)n or one of R2 or R3 is H and the other is Ph; R4= a hydrolytically removable 1-4C alkyl gp., n= 4, 5 or 6. USE/ADVANTAGE - (I) are precursors for aldehydes of formula (VI) which are key cpds. in the synthesis of 3-desmethyl mevalonic acid derivs. which are inhibitors of 3-hydroxy-3-methyl-glutaryl-CoA, reductase and thus influence cholesterol blood levels. (I) can be obtd. in high yields and high optical purity using readily available starting materials and thus provide an economical route to (VI).
