abstract | The prepn. of diaminodiol derivs. of formula (I) and their enantiomers and physiologically acceptable salts comprises treating homochiral alpha-amino-aldehyde derivs. of formula (II) with (V2Cl3(THF)6)2(Zn2Cl6) or a vanadium complex obtd. in situ from VCl3, THF and zinc dust, giving simultaneous control over all 4 centres of durality. In formulae, R1 is a side chain of a naturally or not naturally occurring alpha-amino acid; R1 and R2 = each H or D(E)n(F)o(G)p; E, F and G = each naturally or not naturally occurring aminoacid gps., aza amino acid or imino acid; n, o and p = each 0 or 1; D = R4 or a gp. of formula R4R5N-CR6R7-CO-, R4R8CH-CHR6-CO- or R4O-CHR6-CO- R4 = (a) H, COOH, 1-18C alkyl (opt. mono- or poly unsatd. and opt. substd. with up to 3 opt. different gps. chosen from mercapto, OH, 1-7C alkoxy, carbonoyl, 1-8C alkanoyloxy, carboxy, 1-7C alkoxycarbonyl, F, Cl, Br, I, amino amidino, etc. (b) R4aW (where R4a is as defined for R4 under (a) and W is CO, CS, OCO, SO2, SO, S, NHSO2, NHCO, CH(OH), N(OH) or COV; or where R4 together with R8 and the atoms to which they are attached form a mono or bicyclic, satd. or partially unsatd. ring system with 5-12 ring members, contg. apart from C, opt. one S atom which is opt. oxidised to sulphoxide or sulphone; (c) glycosyl, pref. glucofuranosyl, or glucopyranosyl, etc. (d) an amino protecting gp.
