abstract | New protein-polycation complexes which form soluble complexes with nucleic acids (and their analogues) from human or animal cells are produced using proteins (I) that bind to the cell surface protein expressed by T cells. Pref. the proteins (I) are (esp monoclonal) antibodies or fragments, directed against the cell surface protein, esp. bind to CD4 and contain a gp. 120-binding epitope, esp. from HIV-1 gp. 120 or a homologous protein from a related retrovirus. The polycation is pref. an opt. modified polyamine, an opt. modified histone or a synthetic homologous or heterologous polypeptide, esp. polylysine. The polycation has a charge of 20-500+, and the ratio of protein (I) to polycation is 10:1 to 1:10. New protein (I) nucleic acid complexes are also claimed which may also include a non-covalently bound polycation, esp. one where the nucleic acid inhibits virus, including the replication and expression of HIV-1 and related retroviruses. The nucleic acid is e.g. in the form of a ribozyme opt. with a carrier RNA. |
