abstract | In the quantitative determination of guanine-nucleotide binding proteins (I;G-proteins) in which (I) are bound to membranes of cells and tissue by radioimmune reaction, the new features are (1) the radioligand used is a synthetic peptide homologous to (I) and labelled with 125-iodine, (2) a polyclonal antiserum raised in rabbits is used; (3) purified (I) is used as the standard. The membranes used are pref. completely solubilised by treating with detergent; polyclonal serum is made by immunisation with synthetic peptides or native (I). USE/ADVANTAGE - The method is quantitative; technically simple, rapid (without the ribosylation and gel- electrophoretic sepn. used in known assays); sensitive and accurate. It is useful in studying the regulation of certain cell functions (and of the way in which these are affected by hormones and pharmaceuticals) |
