abstract | 3'-Azido-3'-desoxy-thymidine derivs. (I) are new. R1 = H, alkyl, halogen, trifluoromethyl or halo-alkenyl; R2 = H; or R2 + R4 = -O-; R3 = H; or R2 + R3 + -O-; R4 = H and R5 = azido or R4 = azido and R5 = H; R6 = hydroxymethyl, dibromovinyl, formyl or ethynyl. Prepn. of (I) comprises: (A) oxidn. of (II) with e.g. DMSO or DCCl, followed by a Wittig reaction to give (I) (R6 is -CH=CBr2); (B) debromination of (I) where R6 = -CH=CBr2 to (I) where R6 = -CCH, e.g. using n-BuLi; and (C) for anhydro cpds. (I) where R6 = CH2OH, treatment of corresp. O-protected cpds. with azide salt in DMF/water. USE/ADVANTAGE - Cpds. (I) are useful in treating viral diseases, tumours and bacterial infections. Compared with known nucleosides, cpds. (I) avoid resistance problems, are more selective and have fewer side effects. |
