Fluorophorone-substd. phosphate derivs. (I) F-O-(X)n-R are new: F = fluorescing dye; X = CH2; n = 1-18; R = opt. substd. phosphate, phosphoramidite, H-phsophonate, thiophosphate or phosphoramidate. Fluorescence-labelled cpds. of formula (III) are also claimed. R1 = 3'- or 5'-oligonucleotide, nucleoside or nucleotide. Pref. F is fluorescein or a fluorescein O-hydroxyalkyl ether or a deriv. with a 1-18 CH2 spacer. USE/ADVANTAGE - Cpds. (I) are intermediates for cpds. (III) which are probes and primers in gene technology and molecular biology, esp. in manual and automatic sequence analysis. Cpds. (I) provide a simple route to cpds. (III) in high purity and yields and when used in oligonucleotide syntheses avoid the need for additional anchor groups or special protecting groups as are required e.g. in the processes described in WO 88/00201 and DE 3642939. |
