abstract | A process for determining an analyte (I) in a sample in which a bonding partner (II) for (I) is also present, comprises (a) contacting the sample contg. (I) and (II) with a solid on which (I) or a (I)-analogue is immobilised or with a solid and (I) or a (I)-analogue capable of being immobilised on the solid, so that (I) is released from (II) and (II) is bonded; (b) sepg. at least part of the liq. from the solid; and (c) determining the free (I) in the sepd. liq. A test strip for determining (I) in samples is also new and consists of at least one solid body through which the sample liq. can flow, and on which (I) or the (I)-analogue is immobilised, (ii) at least one solid body through which the sample can flow, and on which a labelled (I) or (I)-analogue or a labelled (II) is impregnated but can be washed out and (iii) at least one solid body through which the sample can flow and on which (II) or (I) is immobilised. USE/ADVANTAGE - The process can be used to determine protein-bound (I), such as vitamin B12, thyroxine and various steroid hormones. The process can determine the total amt. of (I) regardless of whether it is in free or bonded form in the original sample. Sepn. of (II) from the sample is very simple and complete. Any immune or enzyme reactions are not disturbed. |
