abstract | 2',3'-Dideoxyribofuranosides (I) are obtd. by transfer of a 2,-dideoxyribofuranoxyl gp. from a 2',3'-dideoxyribofuranoside (II) to an acceptor base (III) in the presence of Lactobacillus leichmannii nucleoside deoxyribosyl transferase (LL-NDT). (II) is 2',3'-dideoxycytidine. (III) is an opt. substd. purine, pyrazolo(3,4-d)pyrimidine or 8-azapurine base, esp. 4-amino-pyrazolo(3,4-d)pyrimidine, 8-azaguanine, 8-azaadenine, 3-amino-8-azaadenine, a 6-X-purine (where X = H, NH(CH2)6NH2, benzamido, ethoxycarbonylmethylthio, 4-carboxybutylthio or SH), isoguanine, 2-amino-6-methylthiopurine or 6-hydroxy-2-mercaptopurine. The LL-NDT is obtd. by homogenising an L. leichmannii culture broth, centrifuging, chromatographing the supernatant on an anion exchanger, isolating the second eluted enzyme, and purifying this by affinity chromatography on N6-(6-aminohexyl) adenine-Sepharose. |
