abstract |
(A) Prodn. of L-isoleucine (I) is effected by culturing a microorganism under aerobic conditions in an aq. medium contg. DL-alpha-hydroxybutyrate (II) as well as assimilable C and N sources and inorganic salts. The microorganism is a Corynebacterium or Brevibacterium strain which has been grown on a solid medium contg. 0.1-40 mg/ml D-lactate. (B) Mutants of Corynebacterium glutamicum ATCC 13022 or 14310 with increased D-lactate dehydrogenase activity, with the accession nos. DSM 3717 and 3718 respectively are new. A C. glutamicum strain with a D-lactate dehydrogenase activity of 0.05-8 U/mg, esp. DSM 3717 or 3718, is cultured in a medium contg. 20-40 g/l of (II). The medium also contains 0.1-0.25 mg/ml of DL- or L-leucine. |
