abstract | In the quantitative analysis of the concn. of an analyte or of the activity of an enzyme contained in a liq. sample, the reaction velocity is assayed by the photometric determn. of a coloured prod. which is formed in at least 2 steps in the presence of an analyte or enzyme. In the 1st step (1), a correlation equation is set up between an optical density of a coloured prod., which is measured at the moment To after the starting-time of the reaction, and a value of the decrease of the optical density during the time To and a given time T or an average value of the decrease of the optical density per unit time. At least 2 reference solns. are used, contg. an initial cpd. of the reaction for the analysis or an intermediate prod. in a known concn. or activity. (2) The values of the optical density of a coloured prod., prepd. by the effect of the analyte or of the enzyme in the liq. sample at the time to after the starting-time of the reaction and at the time t after the to, are measured. (3) The value of the optical density at the time to is introduced into the correlation-equation above, so as to calculate value of the variation delta at the time t. (4) The concn. of the analyte or the activity of the enzyme in the liq. sample is determined quantitatively by an assay of the reaction velocity. The value delta above is used as correlation-value for varying the optical density between twofold measurement in step (2), to and to + t. USE - The determn. of the concn. or of the activity of a specific component in a liq. sample, e.g. blood, is important in clinical tests. E.g. the enzyme activity value of blood can be used to localise a diseased position and the extent of the illness. The determn. of the concns. of amino-transferase and aspartite-transferase in blood is useful for the diagnosis of liver diseases. |
