Method and reactant for the differentiation of the liver and bone isoenzyme of the alkaline phosphatase and for the selective determination of differentiated isoenzymes by measuring the activity of the alkaline phosphatase, wherein the differentiation of the two izoenzymes is effected by means of a monoclonal antibody directed against one of the two isoenzymes and having only a low cross-reactivity with the other isoenzyme. The present invention also relates to monoclonal antibodies appropriate to the differentiation of the two isoenzymes. Said antibodies are obtained by immunization of test animals with a prepurified alkaline phosphataes from the liver or the bones, fusion of lymphocytes B, cloning and cultivation of hybridane cells thus formed and isolation of the monoclonal antibodies from the latter. |
