abstract | Penicillins of formula (I) including keto tautomers and their salts with bases are new. In (I) A is phenyl (opt. substd. by 4-hydroxy or 3,4-dihydroxy) or 2-thienyl. R is a gp. of formula (A), Q is -SO2.N:CR1R2, -Y.Z.X or (B). R1 and R2 are each methylthio or amino, and when one is amino, tautomeric forms are included. Z' is O, S or :NRz. Rz and R3 are H or 1-3C opt. hydroxy-substd. alkyl. R4 is H, hydroxy, hydroxymethyl or methyl. n is 2-4; when R3 and/or Rz is H, tautomeric forms are included. Y is SO2NH, SO or SO2. Z is 1-3C alkylene. X is hydroxy, aminocarbonyl, aminosulphonyl, formylamino, acetylamino, amino, methylsulphinyl, methylsulphonyl ot -CH(NH2).COOH. (I) are broad-spectrum antibiotics effective against Gram negative and positive species and can be used in human or veterinary medicine for treating or preventing systemic infections. They are also useful as preservatives for polymers, lubricants, dyes, leather, etc. All (I) tested had oral LD50 over 4g. per kg. and subcutaneous LD50 over 2g. per kg. in mice. Usual daily doses are 10-200mg. per day. |
