abstract | In a new procedure for detection of prostaglandins, prostaglandin detection by radio-, fluorescence- and/or enzyme-immunological methods is carried out with the aid of ultrafiltration. The new procedure is applicable to the detection of prostaglandins in tissues, amniotic fluid, plasma and serum. Typically, a 3 ml. portion of the fluid to be examined is placed in a membrane filter with a pore size such that all serum proteins are retained. The ultrafiltrate is a protein-free soln. in which prostaglandins can easily be determined by usual immunological methods. Before or after the ultrafiltration, all fatty acids and interfering substances can be removed by washing with Et2O. Ultrafiltration gives a simple method for sepg. serum proteins from prostaglandins and of thus increasing the sensitivity and accuracy of the determination. |
