Process comprises cooling the specimen to the freezing temp. T(F) by lowering the chamber temp. at a cooling rate B(I); cooling the chamber to a predetermined temp. T(U); holding the chamber at this temp. until the specimen temp. is on the plateous end temp. T(P), leaves the temp. plateau, and falls further; heating the chamber to an intermediate temp. T(O) which is below the specimen temp. T(H); maintaining the chamber at T(O) until a temp. is reached in the specimen at which 85% of the specimen mass is frozen; and lowering the chamber temp. at a cooling rate B(II) until the end temp. required for the specimen is reached. T(U), T(P), T(O), T(F) and B are specified for lymphocytes, stem cells, granulocytes, bone marrow cells, thrombocytes and erythrocytes. |
