abstract | Prostacyclin derivs. of formula (I) and their salts are new. In (I), R1 is H or opt. substd. alkyl; R2 is H or alkyl; R3 and R4 are H or opt. substd. alkyl or aralkyl, provided that one is H when the other is aralkyl; n = 0-2; m = 0-8; A is CN, alkoxycarbonyl, aralkoxycarbonyl, N,N-disubstd. carbamoyl or bicycloalkenyl. Specified activities of (I) are (i) inhibition of blood platelet aggregation, (ii) inhibition of thromboxaneA2 synthesis and antagonism of thromboxaneA2 action, (iii) inhibition of gastric secretion, and (iv) suppression of undesirable gastrointestinal effects due to administration of prostaglandin synthetase inhibitors. Specified uses include treatment of thrombosis, cerebral ischaemic attacks, ulcers, vascular disorders, etc., control of oestrus, and induction of abortion. Cpds. (I) have a narrower activity spectrum than natural prostacyclin, with fewer side effects and a longer-lasting action. |
