abstract | An indirect method for quantitative and qualitative analysis of one partner in an immulogical reaction uses in place of the antibody molecules themselves, their fragments (I) devoid of the Fc portion. (I) are thus unable to fix complement. Other, opt. labelled Fc-fragments can be added which are specific for (I). Pref. an excess of (I) is avoided so that separation of the reaction components is unnecessary. Pref. (I) are F(ab)1 or F(ab)2 fragments and one reaction component can be present in cytoplasm, cell membranes, cell nuclei or tissue. Used for assaying all types of antigens using e.g. the anti-globulin test, the indirect test, with complement or the 'sandwich' test. By removing the Fc portion non-specific binding is minimised so that specificity is improved without loss of the high sensitivity typical of indirect methods. |
