abstract | Prepn. of alkyl- and acylphospholipids of formula (I): (where each R1 is H or 1-4C alkyl; R2 is linear, opt. unsatd. 8-22C alkyl or opt. unsatd. aliphatic or aromatic acid gp.; and R3 is H or CH2OR2) comprises hydrolysing an acetoin dialkyl phosphoric acid ester of formula (II): (where R4 is halogen or -N(R1)3) in presence of an organic solvent and under mild conditions at pH ca. 9-10 and at 20 degrees C. to the b. pt. of the solvent (pref. at 20-60 degrees C.). When R4 is halogen, hydrolysis is carried out in the presence of an amine, N(R1)3. The amino gp. is opt. quaternised with conventional alkylating agents to give a phosphatidyl choline cpd. (I) esp. phosphatidyl choline can be used in the pharmaceutical, cosmetic, food and feedstuffs industries, esp. as pharmaceuticals and emulsifiers. (II) has pref. been prepd. by reacting diacetoinendiol-cyclo pyrophosphate with HO(CH2)2R4 and then with HOCH2CH(OR2)R3 or in the reverse order. |
