titleOfInvention NEW IMMUNO ADSORBENT, METHOD FOR ITS MANUFACTURING AND ITS USE
abstract Immunoadsorbents comprise one of the components (A or B) of a biologically interacting system (A/B) of substances with a natural reversible binding affinity for one another, coupled to an insoluble polymeric carrier by means of a polyfunctional coupling component. The coupling component forms acid-resistant chemical bonds with the reactive gps. of the polymeric carrier and with the biologically active component A or B. Immunoadsorbents comprising immobilised antigens are esp. useful for isolating specific antibodies by affinity chromatography. The immunoadsorbents have high stability and can be re-used repeatedly without severe loss of activity. The A/B system can be an antigen/antibody, enzyme/- inhibitor, hormone/hormone-transporting protein, hormone/antihormone, vitamin/vitamin-binding protein, cell/-anticellular antibody or catalyst/substrate system. The carrier can be a synthetic polymer contg. free NH2, SH, phenolic and/or histidyl gps., esp. a crosslinked copolymer of an aminoalkyl (meth)acrylate with one or more inert comonomers, and the coupling component is pref. 1-3-difluoro-4,6-dinitrobenzene (DFDNB).
