abstract | Pregnan-21-oic acid esters of formula (I): (where ---- is a single or double bond; X is H, F or CH3; Y is H, F or Cl; Z is CH2, beta-hydroxymethylene, beta-acyloxymethylene or, when Y is Cl, also beta-fluoromethylene or beta-chloromethylene; V is CH2, ethylidene or vinylidene; R1 is acyl; and V1 is halogen halogenated 1-5C alkyl or halogenated phenyl), e.g. 11beta-hydroxy-17 alpha-butyryloxy-3, 20-dioxo-4-pregnen-21-oic acid 2-chloroethyl ester, are new cpds. Applied topically, (I) have marked anti-inflammatory activity, but systemically are practically inactive. Onset of action is rapid, and the activity is of high intensity and long duration. In many cases, (I) have good resorption properties, and the cpds. have relatively good stability in formulations. |
