abstract | Mononitrate esters of diols (e.g., 11, 18-dihydroxy-pregna-1, 4-diene-3, 20-dione 18-nitrate) are produced by subjecting a nitrite ester of an alcohol having an H-atom which is conformationally adjacent to the OH gp. or is capable of so being and in which the atoms linking the H-atom and the OH gp. comprise >=2 adjacent atoms which form part of a ring (e.g., pregna-1, 4-dien-11 -ol-3, 20-dione 11-nitrite) to photolysis is the presence of molecular oxygen, forming a corresp. cpd. in which the specified H-atom is replaced by a nitroxy gp. The diol mononitrate prodts. are useful as inters., partic. in the steroid field. The reaction can be used in the prepn. of 19-hydroxy-steroids from 6-hydroxy-steroids, 18-hydroxy-steroids from 11- and 20-hydroxy-steroids, and 32-hydroxy-steroids from 7-hydroxy-steroids. 18-Hydroxy-steroids are useful as inters. for aldosterone derivs. and aldosterone antagonists, and are also effective in hypertension. DELTA1-18-Hydroxy-steroids are useful in the prepn. of tritium-labelled 18-hydroxy-steroids such as 18-hydroxycorticosterone and 18-hydroxy-11-deoxy-corticosterone which are of use in metabolic and diagnostic studies. |
