abstract | A process for isolating a globinolytic enzyme protein material from adult Schistosoma worms esp. of the genus mansoni which may be used as a diagnostic agent for Schistosoma infection, consists in lyophilising the washed worms, homogenising in a buffer, centrifuging, gel filtering through a membrane retaining mols. of M.W. 10,000, concentrating the retained supernatant, chromatographing on Sephadex G-200, eluting and measuring the absorption at 280 nm, and grouping the eluted fractions into 3 groups with peaks eluted at 0.5, 0.8 and 1.0 column vols. of eluate. The second of these groups contains the immunologically active material which is further purified by chromatography on a phenylalanineagarose column and eluting first with Na acetate buffer of pH 3.94 which removes inactive material then with 0.2M-acetic acid of pH 3.0 which elutes the active protein enzyme. This is unstable but may be stabilised with protein stabilisers such as glycerine. The protein molecule contains 246 plus-or-minus 15 amino acid units acid units of 18 different amino acids and has mol. wt. |
