abstract | Cpds. of formula (I): (where R2 is Me, CHO or CH2OH; R1 is OH or (when R2=Me) also H; R3 is H, halogen or lower alkyl; R4 is CN, COOH, alkoxycarbonyl, aryloxycarbonyl or aralkyloxycarbonyl) have positive inotropic activity and are esp. useful as inters. for cardiac glycosides. Cpds. (I) may be prepd. by reacting a cpd. (II): (where R12 is Me or CH2OH; R5 is H, acyl or an easily removable protecting gp.) with a cpd. (III): (R6O)2PO-CHR3R14 (where R14 is selected as R4 but not COOH; R6 (same or different) are 1-4C alkyl) in the presence of a base, pref. alkali metal alcoholate, hydride, amide, hydroxide or carbonate, pref. at 0-20 degrees C, and removing the protecting gp. The product can be further reacted, e.g. by hydrolysis of ester gps., esterification of COOH, or oxidn. of CH2OH to CHO, pref. with CrO3 in pyridine/H2O. |
