abstract | A composition comprising in vitro transcribed RNA obtainable by a method comprising the steps of: A) providing DNA which codes for the RNA; B) in vitro transcription of the DNA in order to obtain a solution which comprises transcribed RNA; andC) diafiltration and / or concentration and / or purification of the solution comprising transcribed RNA by one or more tangential flow filtration (TFF) steps; wherein in step C) the method comprises at least one diafiltration step using TFF; wherein the at least one diafiltration step using TFF in step C) comprises diafiltration with an aqueous salt solution; wherein the aqueous salt solution comprises about 0.1 M alkali metal halide to about 1 M alkali metal halide; wherein the method in step C) comprises at least one further cleaning method before or after the one or more TFF steps; wherein the at least one further purification process is a reverse phase chromatography process; wherein, in the reverse phase chromatography method, a stationary reverse phase and a mixture of an aqueous and an organic solvent is used as the mobile phase for eluting the RNA; and wherein the at least one or more TFF steps comprise using a TFF membrane cartridge. |
