abstract | Sample collection container containing: a blood sample or blood plasma sample; at least one circulating, cell-free, first nucleic acid that is DNA or RNA; a preservative composition containing a preservative selected from diazolidinyl urea, imidazolidinyl urea, dimethoylol-5,5-dimethylhydantoin, dimethylol urea, 2-bromo-2-nitropropane-1,3-diol, oxazolidines, sodium hydroxymethylglycinate, 5-hydroxymethoxymethyl-1-1-aza 3,7-dioxabicyclo [3.3.0] octane, 5-hydroxymethyl-1-1aza-3,7-dioxabicyclo [3.3.0] octane, 5-hydroxypoly [methyleneoxy] -methyl-1-1aza-3,7-dioxabicyclo [3.3 .0] octane, quaternary adamantine or a combination thereof; containing an anticoagulant and a quenching agent; a reaction product formed by the sample and the preservative composition which: (i) is inert to the nucleic acid of the biological sample; and (ii) is free of aldehyde; such that the nucleic acids therein are then suitable for polymerase chain reaction and DNA sequencing without freezing the sample for at least 7 days and the DNA is free of aldehyde-induced (i) nucleic acid-protein crosslinking, (ii) intramolecular and / or intermolecular nucleic acid Nucleic acid crosslinks, or both (i) and (ii), can be obtained by quenching all the free formaldehyde that may be present with the quenching agent from the protectant composition. |
