abstract Cardeolide-genines (I) have a pos. inotropic action on the heart and are intermediates for cardioactive agents. In (I) X=F, Cl, CH3 or lower alkoxy; R2=H, acyl or other easily hydrolysable OH-protective group; R2=OH or OH protected by acyl or other hydrolysable group; R3, R4 (if same)-H, OH or OH protected by acyl or other hydrolysable group; or R4=OH or OH protected by acyl or other hydrolysable group; R2, R3 jointly with 0 in 5-posn gives an orthoester residue. Steroid esters (II) structure (I) with the pyran ring deriv substituent replaced by -CO-CH2-O-CO-CH(X)- -OR5 where substituents have similar meaning to those in (I) but R1 and R4 are not H, are used to prepare (I) by the intramolecular cyclisation of (II).
