abstract | Stable prepns. of formate-dehydrogenase (used for quantitative determination of formate ions) are obtained from cultures of Pseudomonas oxalaticus by preliminarily removing formate ions from the bacteria mass; adding nicotinamide-adenine dinucleotide during the processing of the bacteria; carrying out the decomposition of the cells (by ultrasonic vibrations) in the presence of a buffer maintaining the pH between 5.5 and 6.5; and, finally, destroying the accompanying enzymes (which oxidise dihyro-nicotinamide-adenine dinucleotide) by lowering the pH to 4.5-5.5 by the addition of an acid. |
