abstract The invention relates to immunological methods for the determination of prostate-specific antigen (PSA), in particular for the determination of free PSA, the total content of PSA and the content of PSA-serpin complexes by incubating the sample with at least one antibody which is specific to free PSA, however does not bind to complex PPE. The method is characterized in that an amine-containing nucleophile is added to the sample, in particular before the total PSA content is determined. The invention further relates to the use of amine-containing nucleophiles for the production of free PSA and a method for the production of free PSA from complexed PSA by incubating a sample which contains complexed PSA with an amine-containing nucleophile.
