The invention relates to a method for the amplification of nucleic acid fragments from a sample, which consists of two or three thermocyclic amplification reactions, completely randomized primers are used during the first amplification reaction and specific primers are used in the second amplification reaction, which is characterized in that for amplification the DNA a mixture of at least two DNA polymerases is used, of which at least one polymerase has proofreading activity. In this way, DNA from single cells or cell clones with a low cell count can be used for mutation analysis. |
