Tridecapeptides of formula: Butoc-Gly-Hic-Phe-Arg-Trp-Gly-Lys-Pro-Val-Gly-Lys-Lys-X-NH-(CH2)n- -NH2 (where Butoc is n-butoxycarbonyl, X is lysine or ornithine and n is 2-6) have high adrenocorticotropic activity (ca 90 IU/mg) with the activity spectrum of natural ACTH and are stable at atmospheric oxygen so that they can be stored for prolonged periods without special precautions. They may be prepd by condensing Butoc-Glu(OBut)-His-Phe-Arg-Trp-Gly-OH (where But is tert. butyl) with H-Lys(BOC)-Lys(BOC)-X(BOC)-NH-(CH2)n-NH-BOC (where BOC is tert. butoxycarbonyl) with dicyclohexylcarbodiimide in the presence of activating components the pK of which is 4.0-8.0 and subsequently removing the protecting groups with strong acids such as CF3COOH or HCl. |
